Viral diagnosis in cultivars of Ipomoea batatas (L.) Lam.

Ipomoea batatas (L.) Lam. commonly known as sweet potato, is an important staple food worldwide, mainly due to its high nutritional value and yield. However, vegetative reproduction of potato makes it more susceptible viral infections, which threatens productivity, quality, difficult long-term preservation in germplasm banks. Also, can act a virus reservoir infecting the rest plant accessions bank collections. Hence, this work aimed screen Begomovirus, Potyvirus, Carlavirus infections 16 traditional cultivars from collection ISOPlexis Germplasm Bank, Madeira, Portugal. The infection prevalence by these viruses among was 81.25%, 25.00%, 6.25%, respectively; being ISOP1011 only accession coinfected Potyvirus Carlavirus. ISOP1006, ISOP1010, ISOP1047 were also Begomovirus highlighting their vulnerability infections. ISOP1005 ISOP1027 ones not infected any viruses. analysis partial sequence obtained detected ISOP1011, revealed existence ORF that encodes for 93 amino acids catalytic domain RNA-directed RNA polymerase related Tymovirus protein family, could be confirmed comparison with proteins stored UniProtKB. Multiple alignment showed Motifs A B conserved. search similarity sequences deposited GenBank reported identity Sweet yellow mottle (SPYMV) chlorotic fleck (SPCFV). 9-11% discrepancy nucleotide phylogenetic carried out using maximum probability method suggests isolated new divergent strain SPCFV species.
